HISTO SPOT® 4D Typing Resolution
HISTO SPOT® 4D provides an automated medium resolution typing result for all clinical applications. The typing kits have been designed to uniquely identify common (C) alleles as defined by the Common and Well-Documented Alleles catalogue version: 2.0(1) and generate a result containing a single C allele within a string of other HLA alleles. No string of alleles should contain 2 common alleles at the P group level and there should be no 1st field ambiguities. Typing kits are under continual development to improve resolution and remove ambiguities caused by new published alleles.

HISTO SPOT® Xtend & CWD High Res Typing Resolution
HISTO SPOT® Xtend delivers an increased 4 digit resolution using a supplementary well. The HISTO MATCH software links the additional typing data provided by the Xtend well with the previous HISTO SPOT® 4D analysis to provide a combined higher resolution result.

The HISTO SPOT® CWD High Res typing kit provides a direct to high resolution Class I & Class II solution for a single patient by combining the HISTO SPOT® 4D & HISTO SPOT® Xtend products in a test well holder.

The HISTO MATCH software enables results to be analysed using a common (C) allele & a common and well documented (CWD) allele filter as defined by ref (1). A study was undertaken to show the resolution of the HISTO SPOT® Xtend kits. The study showed that when using the C filter >95% of samples typed were unambiguously resolved (only 2 common alleles found) enabling high resolution typing at the C allele level for P groups.

DNA Extraction Methods
DNA quality and concentration are crucial parameters in all PCR techniques including the HISTO SPOT® SSO system for HLA typing. This study compares the performance of 5 commercially available DNA extraction methods and concludes all are compatible with the HISTO SPOT® SSO System.
